Histochemical skin examination for detection of collagen/elastic fibers (Van Gieson modification)

Description

Histochemical examination – an examination of biopsy or surgical material by a pathologist, prepared as ultra-thin sections and then specially stained. The prepared material is mounted on glass slides and stained with special dyes. A pathologist examines it under a microscope, evaluating the state of cellular structures, their interrelation, the presence of background changes, etc. A histological examination makes it possible to verify pathological changes and to determine the form, stage of the disease, and the background on which it is occurring. One of the main stages of preparing material for histological examination is visualization, which is achieved by staining tissue structures with dyes. Special reagents are applied to the thin sections of material. If a detectable substance is present in the tissue, a chemical reaction occurs between it and the reagent. As a result, specific staining occurs, which is visible under a microscope. Based on the localization and intensity of staining, the pathologist judges the presence, localization, and concentration of the detectable substance. Van Gieson's stain is used to detect elastic fibers in tissue and to assess the degree of fibrosis.
